Needing 142 to make Pakistan bat again, the Proteas were 115 for three, still trailing the visitors by 26 runs.
The day started brilliantly for South Africa when Makhaya Ntini became the third South African and 21st bowler to take 300 runs when he dismissed Mohammad Sami for 10 in the seventh over of the day.
After taking the wicket, a jubilant Ntini raised his shirt to reveal a t-shirt with the handwritten words "Mdingi Express 300 wickets" - Mdingi being the name of the village in the Transkei where he grew up.
Ntini joined Allan Donald and Shaun Pollock as the third South African to achieve this milestone.
